Protein separation and characterization have traditionally been performed by slab gel electrophoresis techniques. There are limitations of 2D-PAGE due to incomplete solubility of hydrophobic and membrane proteins. A major drawback is relatively slow, labor-intensive, and clumsy technology of 2D-PAGE.
A promising high-throughput technique for proteome characterization could be a capillary isoelectric focusing (CIEF) in conjunction with ESI mass spectrometry. The movement form slab gel technique to CIEF technique provides many advantages such as increased speed, improved separation efficiencies, reduced sample volumes, and a capability for automation.
In this seminar, we will demonstrate the effectiveness of CIEF in obtaining high-resolution separations and significant enhanced mass spectral quality for complex protein mixtures with the high sensitivity and mass measurement accuracy of FTICR.
