▶Abctract
Most secreted and membrane-anchored proteins synthesized at the endoplasmic reticulum (ER) contain short cleavable N-terminal hydrophobic signal sequence that varies widely in the primary structure. The signal sequence has been considered only as a sorting signal that mediates specific targeting of client proteins to the endoplasmic reticulum (ER). However, recent studies have uncovered many exceptional regulatory functions of signal sequence; protein translocation, degradation, processing, co-translational folding and topogenesis. Here, we provide a functional rationale for this long-observed sequence diversity by demonstrating that differences among signal sequences facilitate substrate-selective modulation of protein translocation and degradation. This study gives an insight into the therapeutic regulation of signal sequence efficiency as a potential strategy for human diseases associated with proteostasis deficiency.
