Cancer researchers share a common goal: to discover ways to hurt cancer cells without hurting normal cells. My lab has been taking an unusual approach towards this goal: we search for metabolites formed naturally in cells that have toxic properties, and then we examine the processes that produce and break down these toxic metabolites. We have found several instances where the processes that produce such toxic metabolites are upregulated in cancer cells relative to normal cells. In these instances, targeting the enzyme which breaks down the toxic metabolite is only harmful to cancer cells, and not normal cells, as the cancer cells are the only ones in need of detoxification. I will discuss our efforts to identify such pathways and describe the selenocysteine and vitamin B12 metabolic pathways as examples where we have been able to successfully carry out this approach.
