▶Abctract
The phosphorylation of eIF2α upon environmental stress causes not only global translational attenuation but also paradoxically enhanced translation of certain genes. A genome-wide screening using Ribo-seq was performed to identify genes which are preferentially translated upon eIF2α phosphorylation. In contrast to known mechanism for preferential translation, there is no significant difference of uORF occurrence in differentially translated genes, suggesting that other mechanism accounting for this phenomenon. To our surprise, certain genes showed very rapid induction of mRNA amount at transcriptional level through eIF2α phosphorylation as well as translational inhibition. Enhanced transcription of those genes is mediated by the release of paused RNA polymerase II at the promoter proximal region through enhanced p38 kinase activity. When we inhibit the transcription of those immediate early genes in the presence of ER stress, there was markedly decreased cell viability. These findings suggest that translational attenuation by eIF2α phosphorylation causes either preferential translation of certain genes or enhances transcription of same or other genes to respond to the stresses to protect cells from the acute environmental toxicity.
