During vascular development, cambial cells behave as stem cells to produce both xylem and phloem cells. However, vascular tissues are deeply embedded inside the plant body, which makes it difficult to analyze their sequential differentiation process. Previously, our genetic studies revealed that GSK3s play crucial roles in controlling the vascular stem cell differentiation (Ref. 1, 2). By suppressing the GSK3s activity with a chemical compound bikinin, recently we succeeded in the establishment of the tissue culture system VISUAL, in which xylem cells (Ref. 3) and phloem sieve elements (Ref. 4) can be ectopically induced in Arabidopsis leaves. Our transcriptome analysis clearly uncovers co-expression network for phloem-related genes, and thereby illustrating the sequential process of the differentiation (Ref. 4).
In addition to the published results, in this seminar, we would like to introduce our recent efforts to uncover molecular mechanisms underlying the multi-potency of vascular stem cells. Indeed, forward and reverse genetic approaches combined with VISUAL identified key regulators for xylem and phloem differentiation. Furthermore, application of dual-color luciferase reporters into VISUAL enabled the visualization of spatio-temporal dynamics of vascular cell fates.
