Dendritic spines bridge over 90% of neurons in the excitatory network of the brain. How the spines modulate various voltage inputs is critical to understand the information flow of the network. However, the modulatory functions have not been fully explored because of technical limitations. Here, we challenged to reveal the electrical functions by an improved genetically encoded voltage sensor. Back-propagated action potential (bAP) fully penetrated dendritic spines. Excitatory post-synaptic potential (EPSP) propagated from dendritic spine to its parent dendrite was filtered with a factor of 2. In simultaneous inputs of bAP and EPSP, voltage on dendritic spines did not show supra-linear summation. These results show how dendritic spines deal various voltage inputs. Consequently, this sheds light on understanding how dendritic spines regulate the information flow in excitatory network of the brain.
